Ipamorelin, explained simply

A uniquely selective growth hormone booster that tells your pituitary to release more GH without messing with your cortisol or other hormones—making it the 'clean' choice among growth hormone peptides.

What is Ipamorelin?

Ipamorelin is a lab-made peptide that signals the pituitary (the master hormone gland at the base of the brain) gland to release growth hormone in natural pulses. Its distinguishing feature is selectivity: unlike similar peptides, it raises growth hormone without also spiking cortisol (the stress hormone) or other stress hormones. It has been through early-stage human trials, but it is a research compound, not an FDA-approved medicine.

How it works in your body

Think of your pituitary (the master hormone gland at the base of the brain) gland as a GH factory that's controlled by two signals: one that says 'make more!' (GHRH) and one that says 'slow down!' (somatostatin (the hormone that switches growth hormone release off)). Ipamorelin works like pressing the gas pedal without also triggering the alarm system. It binds to the ghrelin (the hunger hormone) receptor (the same one that tells you you're hungry), which signals your pituitary to release growth hormone in natural pulses—just like your body does when you're young. What makes Ipamorelin special is that it ONLY does this one job. Other similar peptides also accidentally boost stress hormones like cortisol (the stress hormone), but Ipamorelin is laser-focused on GH alone.

The deeper mechanism, for the curious

Ipamorelin is a short chain of five amino acids (the building blocks of proteins) that tells your body to release growth hormone (GH). It attaches to the ghrelin (the hunger hormone) receptor, also called GHS-R1a. A receptor is a docking site on a cell. Other GH-releasing drugs, such as natural ghrelin, GHRP-6, GHRP-2 and hexarelin, also raise stress hormones. Ipamorelin does not: it releases GH without significant effects on ACTH, cortisol (the stress hormone), prolactin (a hormone tied to lactation and libido), or aldosterone (hormones involved in stress, milk production, and salt balance). This held at doses more than 200 times higher than the ED50 (the dose that gives half the maximum GH release). Here is how it works, step by step. Ipamorelin attaches to GHS-R1a on the pituitary (the master hormone gland at the base of the brain) cells that make GH. That switches on a relay of signals inside the cell (phospholipase C, then IP3 and DAG), which raises calcium in the cell. The calcium makes the cell empty out the packets that hold GH. GHS-R1a on nerve cells in the hypothalamus (a control center in the brain) may also block the release of somatostatin (a hormone that holds GH back), which boosts the GH response further. In the body, the effect rises in step with the dose. Half of the drug is gone after about 2 hours (terminal half-life (how long it stays active in your body)). Clearance is 0.078 L/h/kg and volume of distribution is 0.22 L/kg (measures of how fast the body removes it and how widely it spreads). The SC50 (the concentration that gives half the maximum GH response) is 214 nmol/L, and the maximum GH production rate is 694 mIU/L/h.

Evidence check

Some human studies

It has been tested in people, but only in small or early-stage studies. Promising, but not yet proven the way an approved medicine is.

Targets Growth Hormone Only

Releases GH without raising ACTH or cortisol (stress hormones) in conscious pigs, holding at doses 200 times above the dose that gives half the maximum GH effect. Never measured in humans. Neither published human study tested for those hormones.

90/100

Recovery Support

No human study has tested ipamorelin for workout recovery, sleep or tissue repair. In the one controlled human trial, trouble sleeping was more common on ipamorelin than on placebo — about 1 in 9 versus about 1 in 19 (10.7% vs 5.2%). The repair evidence is in rats: three months of ipamorelin restored bone formation and muscle strength in rats given a steroid that suppresses both, and it sped gut transit in a rat model of post-surgical ileus.

92/100

Safety Profile

Human exposure is limited to short courses: a single 15-minute IV infusion in the 40-volunteer study of how the body processes the drug, and up to 7 days by IV in the one Phase 2 trial (a mid-stage safety and effectiveness trial), where low potassium, insomnia and high blood sugar were all more frequent than on placebo. There is no data on long-term use, and none on injection under the skin.

What the research points to

  • Makes your pituitary (the master hormone gland at the base of the brain) gland release more of your own growth hormone in gentle pulses that copy your body's natural rhythm
  • Unusually selective: it does not spike cortisol (the stress hormone) or ACTH (stress hormones) the way other growth hormone peptides do, so you avoid stress-hormone side effects
  • Supports muscle recovery and lean body mass gains when combined with exercise
  • May help improve sleep quality, since growth hormone is released during deep sleep
  • Can help counter muscle and bone loss caused by steroid medications
  • Supports fat metabolism indirectly through growth hormone's effects on body composition (the balance of fat and muscle)

The most-asked question

How much Ipamorelin do I take?

The research-reported range is 200 mcg, multiple times daily. Studies used different doses for different situations — find yours:

  1. New users, Assessing tolerance

    100 mcg per injection

    • How often
      2-3 times daily
    • For how long
      4-8 weeks
    • How
      Injection under the skin

Conservative entry dose; Ipamorelin is a selective GH secretagogue (a compound that prompts a hormone's release) with a ~2 hour half-life (how long it stays active in your body), so doses are split through the day 12. Used in research and practice to assess tolerance before titrating (stepping the dose up gradually) up.

  2. Recovery support, GH optimization

    • For how long
      8-12 weeks
    • How
      Injection under the skin

Most commonly cited research/practice dose; gives a meaningful pulsatile GH release without raising cortisol (the stress hormone) or prolactin (a hormone tied to lactation and libido) at standard doses 12.

  3. Experienced users, Maximizing GH pulse

    300 mcg per injection

    • How often

Adjusting your dose

You're at the right dose if…

  • Improved sleep quality—waking up more refreshed
  • Faster recovery between workouts
  • Gradual improvements in body composition (less fat, more muscle tone)
  • Better skin quality and overall sense of vitality

Consider going up if…

  • You've used the starting dose for 2+ weeks with zero side effects
  • You're not noticing any changes in sleep, recovery, or body composition
  • You're stacking with a GHRH and want amplified effects

Go down if…

  • You experience persistent headaches that don't resolve
  • Water retention becomes uncomfortable
  • You notice tingling in hands that worsens (carpal tunnel signs)
  • Blood sugar becomes harder to control

The ceiling: 300 mcg three times daily (900 mcg total). Higher doses haven't shown proportionally greater benefits in research.

Reconstitution, in plain words

How do I mix Ipamorelin?

Peptides usually arrive as a freeze-dried powder in a small vial. “Reconstitution” just means adding sterile water back in so you can measure a dose. Here's how it works for Ipamorelin:

What you need

  • Bacteriostatic water (BAC water)—the preservative allows multiple uses
  • Insulin syringes (29-31 gauge, 0.5mL or 1mL)
  • Alcohol swabs for cleaning vial tops and injection sites
  • Your Ipamorelin powder vial (typically 2mg or 5mg)

A worked example

For a 5mg vial: Add 2.5mL of bacteriostatic water to get a concentration of 2mg/mL (2000 mcg/mL). Each 0.1mL (10 units on an insulin syringe) equals 200 mcg of Ipamorelin.

Working out your dose

At 2mg/mL concentration: For a 200 mcg dose, draw 0.1mL (10 units). For 300 mcg, draw 0.15mL (15 units). For 100 mcg, draw 0.05mL (5 units).

Or let the calculator do it

Three quick questions — your vial, your water, your dose — and it tells you the exact syringe mark. “Find dose amount” is preloaded with Ipamorelin's recorded schedule.

Injection sites & technique

Where do I inject Ipamorelin?

The best spots

  • Belly fat area (about 2 inches from your belly button)
  • Front or outer thigh (middle section)
  • Back of the upper arm (pinch the skin if needed)

How to do it, step by step

  • Wash your hands thoroughly with soap and water
  • Clean the injection site with an alcohol swab and let it air dry
  • Pinch about an inch of skin to create a fold
  • Insert the needle at a 45-90 degree angle (45 for lean individuals, 90 if you have more tissue)
  • Push the plunger slowly and steadily over 5-10 seconds
  • Wait 5 seconds before removing the needle to ensure full delivery
  • Apply light pressure with a cotton ball if needed—don't rub the site

Subcutaneous (under the skin) injection (just under the skin)—quick, easy, and most effective for peptides like Ipamorelin

Timing

When do I take Ipamorelin?

Best time of day

Inject on an empty stomach—ideally first thing in the morning, post-workout, and before bed. The bedtime dose aligns with your natural overnight GH surge. Wait at least 30 minutes before eating after injection.

With or without food?

Food, especially carbs and fats, can blunt the GH response. Always inject at least 30-60 minutes before meals or 2-3 hours after eating for best results.

Taking it with other peptides

When pairing with CJC-1295, inject them together at the same time—they work synergistically. If using BPC-157 or TB-500 for healing, you can inject those at different sites or times.

The honest list

What side effects could Ipamorelin cause?

Common — usually mild

These show up most often. They usually fade as your body adjusts.

  • Mild headache

Some users experience headaches in the first few days to weeks. This usually relates to your body adjusting to increased GH pulses.

What to do Stay well-hydrated, start with lower doses, and it typically resolves within the first 1-2 weeks.

  • Injection site reactions

Redness, minor swelling, or itching at the injection site is normal and harmless.

What to do Rotate injection sites between belly, thigh, and arm. A cool compress can help. Usually fades within hours.

  • Water retention

Mild bloating or puffiness, especially in the first few weeks. This is a known GH effect that usually stabilizes.

What to do Reduce sodium intake, stay hydrated, and give your body 2-3 weeks to adjust. Consider slightly lower doses if significant.

  • Increased appetite

Ipamorelin activates the ghrelin (the hunger hormone) receptor, which can stimulate hunger—the same receptor that makes your stomach growl.

What to do Plan your meals around injection times. This effect can actually be useful if you're trying to gain muscle and need to eat more.

Less common

Worth knowing about, but most people never see these.

  • Tingling in hands or feet

GH can cause temporary fluid changes that press on nerves, creating carpal tunnel-like symptoms.

What to do Usually mild and temporary. If persistent, reduce dose. Wearing wrist splints at night can help.

  • Joint stiffness

Some users notice temporary joint discomfort as GH affects connective tissues and fluid balance.

What to do Stay active with gentle movement, stay hydrated. Usually resolves as your body adapts over 2-4 weeks.

Serious — stop and get medical help

Rare, but if any of these happen, stop and contact a medical professional.

  • Allergic reaction

Rare but possible with any peptide. Watch for hives, significant swelling (especially face/throat), or difficulty breathing.

What to do STOP immediately and seek emergency medical care. This is extremely rare but requires immediate attention.

Not seeing anything? The usual reasons

  • Eating too close to injection—food, especially carbs, blunts GH release. Always inject fasted.
  • Inconsistent dosing schedule—GH peptides work best with consistent timing to establish rhythmic release
  • Poor quality peptide from unreliable source—always verify third-party testing certificates (COAs)
  • Improper storage degraded the peptide—check that solution is crystal clear before using
  • Unrealistic expectations—Ipamorelin provides gradual, natural improvements, not overnight transformations
  • Not supporting with proper nutrition and exercise—peptides enhance your lifestyle, they don't replace it
  • Blood sugar too high at injection time—insulin spikes inhibit GH release

Is it right for you?

Who should avoid Ipamorelin?

Do not use it if…

  • You have active cancer or a history of cancer—GH can potentially fuel tumor growth
  • You are pregnant or might become pregnant—effects on fetal development are unknown
  • You are breastfeeding—peptide could potentially pass into breast milk
  • You have a pituitary (the master hormone gland at the base of the brain) tumor or disorder—could worsen the condition
  • You have uncontrolled diabetes—GH can affect blood sugar regulation
  • You're under 18—growth plates are still active and shouldn't be manipulated

Be extra careful if…

  • You have diabetes or prediabetes—monitor blood sugar closely as GH affects glucose
  • You're on blood pressure medications—some users experience changes
  • You have carpal tunnel syndrome—GH can worsen symptoms temporarily
  • You take thyroid medications—GH may affect thyroid hormone conversion
  • You have a history of joint problems—GH can initially cause joint discomfort
  • You're over 65—start with lower doses and monitor carefully

When to stop

  • Any signs of allergic reaction—stop immediately and seek medical help
  • Persistent carpal tunnel symptoms that worsen despite dose reduction
  • Significant joint pain that interferes with daily activities
  • Blood sugar control becomes significantly harder to manage
  • Unusual swelling that doesn't resolve with lower doses
  • Any side effect that feels serious or really concerns you

What it mixes badly (and well) with

Medications

  • Avoid
    Insulin

GH can change how your body responds to insulin. People with diabetes need to check blood sugar closely and may need insulin changes. Talk to your doctor.

  • Avoid
    Diabetes medications (metformin, etc.)

GH changes how the body handles sugar. Check your blood sugar more often and tell your doctor.

  • Check first
    Thyroid medications

GH can change how the thyroid hormone T4 is converted into its active form, T3. You may need thyroid level checks if you take thyroid replacement.

  • OK together
    Glucocorticoids (prednisone, etc.)

Possibly helpful: research shows Ipamorelin can help counter steroid-induced muscle and bone loss. Talk to your doctor.

  • Check first
    Blood pressure medications

Some users experience BP changes with GH peptides. Monitor blood pressure regularly.

Other peptides

  • OK together
    CJC-1295 (with or without DAC)

A very good pairing. CJC-1295 supplies the GHRH signal (the brain's natural growth-hormone-releasing message) while Ipamorelin boosts it through the ghrelin (the hunger hormone) receptor (the docking site for the hunger hormone). Inject together for best results.

  • OK together
    Sermorelin

Another great pairing. Both trigger growth hormone (GH) release but in different ways, creating a stronger natural GH pulse.

  • OK together
    BPC-157

Safe to combine for better recovery. BPC-157 handles tissue repair while Ipamorelin raises overall GH for body-wide recovery.

  • OK together
    TB-500

Complementary for healing. Inject at different sites but can use on the same day.

  • OK together
    GHRP-6

Usually redundant since both attach to the ghrelin (the hunger hormone) receptor. Ipamorelin is preferred because it targets GH more precisely (selectivity). Using both together is unnecessary.

Supplements

  • Check first
    Arginine

Can enhance GH release when taken separately. Some people take arginine at different times to compound effects.

  • OK together
    GABA

Safe to combine—GABA may support additional GH release, especially when taken before bed.

  • OK together
    Melatonin

Safe and potentially synergistic for sleep quality and nighttime GH release.

  • Check first
    High-dose glucose or carbs

Blood sugar spikes blunt GH release. Avoid carbs around injection times for best results.

Who it tends to suit

People Wanting a Clean Growth Hormone Boost

If you want the benefits of increased growth hormone without spiking cortisol (the stress hormone) or other stress hormones, Ipamorelin is uniquely selective. Unlike GHRP-2 or GHRP-6, it won't raise your cortisol levels even at doses 200 times higher than needed for GH release.

Recovery-Focused Athletes

Ipamorelin shines for athletes who want better recovery between training sessions. The GH boost helps repair muscle tissue faster, and since it doesn't mess with other hormones, you can use it consistently without the rollercoaster effects.

Anti-Aging Enthusiasts

Growth hormone naturally declines as we age, contributing to muscle loss, fat gain, and reduced vitality. Ipamorelin offers a gentle way to restore more youthful GH levels without the risks of actual HGH therapy.

Those Recovering from Steroid Use

Research shows Ipamorelin can help counteract the muscle and bone-weakening effects of glucocorticoid medications. If you've been on steroids for medical reasons, this peptide may help you rebuild what you've lost.

The safety picture overall

Ipamorelin's main selling point is that it raises growth hormone without raising cortisol (the stress hormone) or ACTH, two stress hormones that similar peptides do raise. That was shown in pigs, and it held even at doses more than 200 times higher than the amount needed to raise growth hormone 1. It has not been checked in people. The two published human studies measured ipamorelin and growth hormone only. Neither one tested for cortisol, ACTH or prolactin (a hormone that affects milk production and sex drive) 23.

Most of what we know about safety in people comes from a single trial in patients recovering from bowel surgery. 114 people took part: 56 got ipamorelin through an IV for up to seven days, and 58 got a placebo. Most side effects were mild to moderate, and were the kind you would expect after surgery 3.

Serious problems happened at about the same rate in both groups, roughly 1 in 6 either way (17.9% on ipamorelin, 15.5% on placebo). But three things were more common on ipamorelin: low potassium in about 1 in 8 people versus about 1 in 30 on placebo (12.5% vs 3.4%), trouble sleeping in about 1 in 9 versus about 1 in 19 (10.7% vs 5.2%), and high blood sugar at discharge in about 1 in 7 versus about 1 in 12 (14.3% vs 8.6%) 6.

Two people taking ipamorelin died. Both had colon cancer surgery, and in both cases the surgical join in the bowel leaked afterward. Nobody on the placebo died. The FDA says it is unclear whether those deaths had anything to do with ipamorelin. But it pointed to them, along with the potassium and blood sugar differences, when it recommended in October 2024 against letting pharmacies compound ipamorelin 6.

Ipamorelin has been tested in people more than most research peptides, and far less than an approved medicine. There is early work measuring how it moves through the body: 40 healthy men across five dose levels, given by IV infusion, which is where the roughly 2-hour half-life (how long it stays active in your body) comes from 2. There is also the one trial in surgery patients 3. That trial did not work. It was built to see whether ipamorelin got the gut moving again faster after surgery. People on ipamorelin ate their first solid meal about 7 hours sooner (25.3 hours versus 32.6 hours), but that difference was small enough that it could easily have been chance, and none of the other measures separated from placebo either. The company stopped developing it for that use 3. The 1998 study behind the "selective" label was done in rat pituitary (the master hormone gland at the base of the brain) cells (cells from the hormone control gland at the base of the brain), in rats, and in pigs, not in people 1. Ipamorelin is not approved for anything, and in October 2024 the FDA recommended against adding it to the list of substances pharmacies are allowed to compound 6.

Keeping it good

How do I store Ipamorelin?

Before mixing (powder)

Keep your Ipamorelin powder in the refrigerator (36-46°F / 2-8°C) for regular storage. For long-term storage over 1 month, keep frozen at -4°F (-20°C). Store in original sealed vial away from light. Properly stored powder remains stable for 12-24 months.

After mixing (liquid)

Once mixed with bacteriostatic water, refrigerate at 36-46°F (2-8°C). Never freeze the reconstituted solution as this destroys the peptide. Keep away from light and use within 28-30 days for optimal potency.

Signs it has gone bad

  • Cloudy or hazy appearance (should be crystal clear)
  • Visible particles floating or settled at the bottom
  • Color changes—any yellowing or discoloration means it's degraded
  • Reduced effectiveness after proper dosing (peptide may have broken down)

Not our opinion — the published record

What the studies actually found about Ipamorelin

1998

Ipamorelin, the first selective growth hormone secretagogue

This Novo Nordisk study introduced ipamorelin as the first growth hormone (GH) releaser that targets GH only. Potency was measured in rat pituitary cells (cells from the gland that makes GH) and in rats. Selectivity was tested in conscious pigs. Unlike GHRP-6 and GHRP-2, ipamorelin did not raise ACTH or cortisol (stress hormones) above the levels seen with GHRH (the brain's own GH signal). That held at doses more than 200 times above the ED50 (the dose that gives half the maximum GH release). No human study has since measured ACTH, cortisol or prolactin after ipamorelin, so the selectivity finding is still animal data.

Raun K, Hansen BS, Johansen NL, et al.

2014

Prospective, randomized, controlled, proof-of-concept study of the Ghrelin mimetic ipamorelin for the management of postoperative ileus in bowel resection patients

This Phase 2 proof-of-concept trial (an early test of whether a drug works) randomized 114 bowel-resection patients (people who had part of their bowel removed): 56 got ipamorelin and 58 got placebo, into a vein twice daily for up to 7 days. It did not meet its main goal. The median time to first tolerated meal was 25.3 hours on ipamorelin vs 32.6 hours on placebo. That 7.3-hour gap was not statistically significant (it could easily be chance), and the secondary goals also showed no significant difference. Most adverse events were mild to moderate and were blamed on the surgery. Serious adverse events occurred in 17.9% of ipamorelin patients vs 15.5% on placebo. Hypokalemia (low potassium) (12.5% vs 3.4%), insomnia (10.7% vs 5.2%) and hyperglycemia (high blood sugar) at discharge (14.3% vs 8.6%) were more common on ipamorelin. Two ipamorelin-treated patients died after an anastomotic leak (a leak where the bowel was rejoined). FDA's 2024 review states it is unclear whether those deaths were related to ipamorelin. The authors advised reading the results with caution because the sample was small.

2024

Ipamorelin: FDA Pharmacy Compounding Advisory Committee briefing document

FDA's review of ipamorelin before its 29 October 2024 advisory committee meeting. It gives the adverse event rates for each group in the Phase 2 bowel-resection trial, which the journal abstract does not: serious adverse events in 10 of 56 on ipamorelin (17.9%) versus 9 of 58 on placebo (15.5%); hypokalemia (low potassium) 12.5% versus 3.4%; insomnia 10.7% versus 5.2%; hyperglycemia (high blood sugar) at discharge 14.3% versus 8.6%. Two ipamorelin-treated patients died of complications after an anastomotic leak (a leak where the bowel was rejoined) following colon cancer surgery, and none on placebo. FDA states "it is unclear whether the two deaths were related to ipamorelin." It cited the deaths alongside the potassium and sugar imbalances when it recommended against adding ipamorelin to the 503A compounding list (the list of drugs pharmacies may make in bulk).

Your questions

Ipamorelin FAQ

What makes Ipamorelin different from other growth hormone (GH) peptides like GHRP-6?

Ipamorelin is unusually selective. It releases growth hormone without raising cortisol (a stress hormone), ACTH (the signal that tells your body to make cortisol), or prolactin (a hormone that affects milk production and sex drive), as other peptides do. Even at doses 200 times higher than needed for GH release, it doesn't affect these other hormones. That makes it the "cleanest" choice, with fewer side effects and less hormonal disruption.

How long does it take to see results from Ipamorelin?

Most people notice improved sleep quality within the first 1-2 weeks. Recovery benefits typically become apparent by weeks 3-4. Body composition changes (fat loss, muscle improvements) usually take 6-12 weeks of consistent use combined with proper diet and exercise. It's a gradual process, not an overnight transformation.

Why do I need to inject multiple times per day?

Ipamorelin has a short half-life (how long it stays active in your body) of about 2 hours, meaning it clears your system quickly. Multiple injections throughout the day create multiple GH pulses, mimicking how your body naturally releases growth hormone. This pulsatile pattern is more effective and natural than one big dose.

Can I take Ipamorelin with food?

You should NOT take it with food. Eating—especially carbohydrates—causes insulin release, which blunts your GH response. Always inject on an empty stomach (ideally first thing in morning, 2+ hours after meals, or before bed) and wait at least 30 minutes before eating.

Is Ipamorelin the same as HGH (human growth hormone)?

No, they're quite different. HGH (human growth hormone) is the actual hormone injected directly. Ipamorelin signals your own pituitary (the master hormone gland at the base of the brain) gland to release more of YOUR natural GH. This creates a more natural pulsatile release pattern and lets your body maintain its feedback systems. It's gentler and generally has fewer side effects than exogenous (supplied from outside the body) HGH.

Should I combine Ipamorelin with CJC-1295?

Many people do, and it's a popular combination. CJC-1295 mimics GHRH (the hormone that tells your pituitary (the master hormone gland at the base of the brain) to make GH), while Ipamorelin works through the ghrelin (the hunger hormone) receptor. Together, they amplify each other's effects through two different pathways. You can inject them at the same time for convenience.

Will Ipamorelin make me hungry all the time?

It can increase appetite since it activates the same receptor that makes you feel hungry (the ghrelin (the hunger hormone) receptor). However, this effect is usually mild and some people find it helpful if they're trying to eat more for muscle building. If hunger is problematic, it typically becomes less noticeable after the first few weeks.

Is Ipamorelin legal?

Ipamorelin is not FDA approved and is classified as a research compound. It's legal to purchase for research purposes in many countries, but it's not approved for human medical use. Athletes should note that it's banned by WADA and most sports organizations. Always check your local regulations.
